摘要
【目的】研究灰胡杨的发芽周期和种子活力情况。【方法】在常温干燥条件下不同储藏时间对灰胡杨种子发芽率和相关酶的影响,NaCl与PEG6000对灰胡杨种子发芽率的影响;不同储藏时间的灰胡杨种子;用比色法测定种子酶活性,滤纸法测定发芽率。【结果】NaCl、PEG及交互作用对种子发芽率有极显著影响,NaCl作用最大;10%的PEG-6000处理储藏到第27、37 d灰胡杨种子,能够提高种子发芽率。随储藏时间延长,灰胡杨种子发芽率和含水量下降,含水量从第0 d到第37 d含水量范围在6.5%~4.6%,发芽率在第0 d到第37 d从66.7%下降至13.3%。灰胡杨发芽后种子ACP酶活性高峰期出现在储藏第0 d,ACP含量在1.85 mol/(min·g),第27 d酶活性下降较明显。第37 d时ACP含量下降为0.54 mol/(min·g)。灰胡杨种子脱氢酶活性随储藏时间延长呈下降趋势,TTC活性在20.1 mg/mL,储藏到第37 d时TTC活性下降至4.0mg/mL。灰胡杨种子浸泡液电导率显著上升,电导率值变化范围为:113.75~160.94 us/(cm·g)。【结论】通过多元回归分析,灰叶胡杨种子绝对含水量与ACP、TTC活性及种子发芽率呈正相关,与电导率值呈负相关。
[ Objective ] The purpose of the experiment was to determine the characteristics of Populus pruinosa seed germination. [ Method ] The effects of different storage times of Populus pruinosa at normal temperature and NaCl & PEG6000 on the germination rate and correlative enzyme were researched in the project. Colorimetry was used to measure the enzymic activity of seed and filter paper method to measure the germination rate. [ Result ] The results were as follows: the interaction of NaCI and PEG had a significant effect on seed germination rate. NaC played a more important role than interaction in seed germination rate. The best treated Populus pruinosa seed was 10% PEG - 6000 which was higher than other concentration PEG - 6000 treatment. When Populus prt, inosa seed has been stored till 27 and 37 days, 10% PEG -6000 played a role of physiology and biochemistry metabolism and repairing on seed. 10% PEG -6000 could improve of resistance and germination rate. As the seed has been stored for longer time, the germination rate would be in gradual decline as well as water content. From 0 to 37 days of the storage, water content would range from 6.5% -4.6% and the germination rate descended from 66.7% to 13.3%. The peak of the ACP enzyme activity was 1.85 mol/( min·g) on the 0 day of the storage, while on the 17th day it would embrace a stable decline in ACP enzyme activity and on the day 27 sharp decline in enzyme activity. On the day 37, ACP content decreased to 0.54 tool/( min·g). The dehydrogenase activity will be on a declining curve as the storing days increase. The first 7 days will be a high level of activity with Trc activity range from 20.1 - 17.2 mg/mL and the activity fall sharply fall to 4.0 mg/mL TTC activity on day 37. As the storing period increasing, the seed vigor will be in decline and the conductivity of the Populus pruinosa soak solution will go up with an conductivity range of 113.75 - 160.94 us/( cm·g). [ Conclusion] As a result of multiple regression analysis, the water content of the seed has a positive relation with ACP, TTC activity, conductivity and seed germination rate and a negative relation with conductivity data.
