摘要
目的:观察细胞因子TNF-α、TGF-β在胶原2佐剂型关节炎(CIA)大鼠肺组织中的表达,探讨RA继发性肺损害的相关生物学因素。方法:将16只大鼠随机分为生理组、模型组各8只,向模型组动物尾根部皮内注射Ⅱ型胶原(CⅡ)和不完全弗氏佐剂(IFA)混合乳化液0.2 mL致炎,制造类风湿关节炎CIA动物模型,致炎4周后,动物取材,计算肺系数,用酶联免疫吸附试验(ELISA)分别检测大鼠肺组织匀浆液细胞因子TNF-α、TGF-β含量,用电镜观察各组大鼠肺组织形态学变化。应用SPSS 17.0统计软件分析检测结果。结果:CIA模型组大鼠肺系数显著升高,肺组织匀浆液中细胞因子TNF-α、TGF-β含量水平明显升高,与生理组比较差异有统计学意义(P<0.05)。结论:细胞因子TNF-α、TGF-β可能参与了RA发病,同时与RA继发肺损害密切相关。
Objective: To observe the expression of cell factors TNF -α, TGF - β in the lung tissue of rats with collagen 2 adjuvant arthritis, and to explore the relevant biological factors of RA secondary lung damage. Method : 16 rats were randomly divided into normal group and model group, 8 rats in each group. Intradermal injection of collagen type II ( C II ) and incomplete Freund,s adjuvant ( IFA ) hybrid emulsion 0. 2mL were given to tail root of rats to make animal model with CIA rheumatoid arthritis. Four weeks after the inflammation, the lung coefficient was calculated, enzyme-linked immunosorbent assay ( ELISA ) was used to detect the homogenate cytokine TNF -α, TGF - β content of the rat lung tissue, electron microscopy was used to detect the morphological changes of rat lung tissues in each group. SPSS 17.0 statistical software was applied to analyze test results. Re- sults : The lung coefficient of CIA model group was significantly increased, the cytokine TNF -α, TGF - β levels of pulmonary tissue homogenate were significantly elevated, and compared with those of the physiological groups there was a significant difference(P 〈0.05 ). Conclusion: The cytokine TNF - α, TGF - β may be involved in the pathogenesis of RA, and may be closely related to RA secondary lung damage.
