摘要
目的研究用于肿瘤磁流体热疗的超顺磁性氧化铁纳米粒(以下简称纳米粒)的制备、表征及其生物相容性,为其临床应用奠定基础。方法采用改良的化学共沉淀法制备纳米粒。采用透射电镜(TEM)、X射线粉末衍射(XRD)、能谱分析(SEM-EDS)、振动样品磁强计(VSM)等手段对其形貌、物象、成份、磁性能等表征进行观察;MTT试验评价其体外细胞毒性;溶血试验评价其溶血作用;小鼠腹腔注射纳米粒混悬液测定其急性毒性;微核试验评价其致畸、致突变作用。结果相关检测结果显示,纳米粒形态规则、密度均匀,粒径约为23 nm;其成分为Fe与O两种元素。纳米粒对L-929细胞的毒性为0~Ⅰ级;无溶血作用及致畸、致突变作用。结论本研究成功制备了纳米粒;该纳米粒具有良好的生物相容性,在肿瘤热疗领域具有一定应用前景。
Objective To evaluate the preparation, characterization and biocompatibility of Fe3O4 magnetic mnanop- articles for the treatment of tumor by magnetohydrodynamic heat therapy, so to lay the foundations for its application. Methods Fe3O4 magnetic nanoparticles were prepared through co-precipitation method. The appearance, phase constitue, function groups and magnetic susceptibility were characterized by transmission electron microscope (TEM) , X-RAY powder diffraction (XRD), scanning electron microscope-energy dispersive spectrum (SEM-EDS) and vibrating sample magnetom- eter(VSM) ; MTT assay was used to investigate the in vitro toxicity and hemolysis test was carried out to estimate whether it had blood toxicity; Fe3O4 suspension was intraperitoneally injected into KM mouse to calculate the acute toxicity; micronu- cleus (MN) were reckoned to identify whether it was genotoxic. Results TEM showed the shapes of Fe3 O4 nanoparticles to be approximately global and the size was estimated to be about 23 nm ; XRD comparing with powder diffraction file of spinel Fe3 04 nanomaterials by JCPDS indicated only spinel pHase ; The components of O and Fe were determined ; the toxicity to L-929 eel1 was between 0-1 degree and it had no hemolysis activity; No genotoxic effects were found. Conclusion The serf-prepared nanosized Fe3 04 is a kind of high bioeompatibility materials and is suitable for further application in tumor hy- perthermia.
