摘要
目的:探讨醋柳黄酮对心肌缺血再灌注家兔心肌组织中Bcl-2及Bax蛋白及基因表达变化的作用,揭示醋柳黄酮治疗心肌缺血再灌注损伤作用机制。方法:采用结扎家兔冠状动脉前降支方法制备急性心肌缺血再灌注模型,west ern bl ot及r eal-t i me PCR方法检测Bcl-2及Bax蛋白及基因表达变化。结果:与假手术组比较,模型组心肌缺血再灌注家兔心肌组织中Bcl-2基因表达上调2.52倍,Bax基因表达上调5.02倍;经醋柳黄酮治疗后,与模型组比较,心肌缺血再灌注家兔心肌组织中bcl-2基因表达上调1.77倍,bax基因表达下调2.50倍。结论:醋柳黄酮可以通过促进抗凋亡基因Bcl-2基因及蛋白表达,并抑制促凋亡基因Bax基因及蛋白表达而对心肌缺血再灌注起保护作用。
Objective Studied the effect of bcl-2 and bax of total flavones on rabbit with myocardial ischemia and reperfusion, revealed the mechanism of the total flavones on treating myocardial ischemia and reperfusion.Methods Ligated the anterior descending coronary artery of rabbit to result the model of myocardial ischemia and reperfusion, western blot and real-time PCR detected the expression of bcl-2 and bax.Results Compared with the model group, expression of bcl-2 is rised by total flavones treated,but bax is reduced. Conclusion Total flavones can rise the expression of bcl-2 and reduce the expression of bax to protect the myocardial organization with myocardial ischemia and reperfusion.
