摘要
目的 :研究PCR SSCP技术检测原发性胃腺癌石蜡标本中p53第 7外显子基因突变 ,探讨其与肿瘤发展的关系。方法 :应用PCR SSCP(聚合酶链反应 单链构象多态性 )技术检测 37例原发性胃腺癌标本中p53第 7外显子的基因突变 ,同时应用免疫组织化学技术检测p53蛋白的表达。探讨其与肿瘤发展的关系。结果 :37例原发性胃腺癌标本中 ,p53第 7外显子基因突变率为 19% (7/ 37) ,7例发生突变的标本 ,其p53蛋白表达亦呈阳性 ,低分化腺癌突变率高于高、中分化腺癌 (P =0 0 0 0 6 ) ,发生淋巴结转移组突变率高于未发生淋巴结转移组 (P =0 0 0 0 3)。结论 :临床检测胃腺癌原发灶中是否存在p53第 7外显子基因突变 ,有助于识别高度恶性的胃腺癌。
Objective:To investigate the mutation of p53 gene in exon 7 and expression of p53 protein in primary gastric adenocarcinomas, and to study its relation with tumor′s development Methods:The mutation of p53 gene in exon 7 in 37 cases of primary gastric adenocarcinomas were detected by polymerase chain reaction single strand conformation polymorphism(PCR SSCP) and the expression of p53 protein in 37 cases of primary gastric adenocarcinomas was analyzed by immunohistochemistry Results:In 37 cases of primary gastric adenocarcinomas,7cases (19%) showed mutation of p53 gene in exon 7 and positive p53 expression.The mutational rate of p53 gene in exon 7 and positive p53 expression.The mutational rate of poorly differentiated adenocarcinomas was higher than that of well/moderately differentiated ones( P =0 000 6).The mutational rate of those with lymph node metastasis was higer than that of those without lymph node mdtastasis( P =0 000 3).Conclusions:It is useful to recognize highly malignant primary gastric adenocarcinomas by detecting the mutation of p53 gene in exon 7
