摘要 目的:探讨苦参碱对HepG2细胞代谢水平及基因水平的影响。方法:使用不同浓度的苦参碱作用HepG2细胞3天,对PCNA、AFP、N-ar s、e-myc、p53、Rb进行免疫组化检测,对e-myc、p53 mRNA进行原位杂交法检测,对HSGGT进行亲和微柱法检测。结果:苦参碱用药3天后,苦参碱0.8g/L时HepG2细胞AFP和PCNA表达有所减弱,用药1.5g/L时HepG2细胞AFP和PCNA表达为阴性;用药后HepG2细胞相关代谢指标HSGGT明显活性下降,癌基因e-myc表达减弱,抑癌基因p53、Rb表达增强。结论:苦参碱具有抑制人肝癌HepG2细胞增殖的作用,能影响HepG2细胞的增殖代谢及抑制癌基因的增殖。 Objective To study the Effect of matrine on the metabolic level and the level of gene in HepG2 cells, Methods HepG2 cells were cultured about 3 days in different concentrations of matrine, PCNA, AFP, N-ars, e-myc, p53, Rb were detected by immunohistochemistry, e-myc, p53 mRNA by sttu hybridization assay, HSGGT by pro-and micro-column method, Results After 3d,at 0.8g / L of matrine, AFP and PCNA expression weakened at 1.5g / L,AFP and PCNA were negative; The activity of HSGGT decreased obviously, expression of the oncogene of the e-myc decreased,expression of tumor suppressor gene p53 and Rb increased. Conclusion Matrine can affect the proliferation of HepG2 cells metabolism and inhibit the proliferation of cancer genes.
