卡维地洛对肝星状细胞增殖与凋亡的影响 被引量:1

摘要 目的观察卡维地洛对肝星状细胞增殖与凋亡的影响。方法以不同剂量卡维地洛(CVD)作用于肝星状细胞(HSC-T6)。CCK-8法检测CVD对细胞增殖的影响,流式细胞仪检测CVD对细胞凋亡的影响。统计学处理采用t检验。结果 CVD作用于HSC-T6后,CCK-8法检测模拟对照组细胞增殖抑制率为2.18±0.60,CVD10μmol/L组细胞增殖抑制率为6.30±1.44,CVD20μmol/L组细胞增殖抑制率为7.75±1.33。提示细胞增殖抑制率随CVD浓度的增高而增高。CVD10μmol/L组及CVD20μmol/L组与模拟对照组相比,差异有统计学意义。流式细胞术检测模拟对照组细胞凋亡率为(2.30±0.08)%,CVD10μmol/L组细胞凋亡率为(3.54±0.50)%,CVD20μmol/L组细胞凋亡率为(4.25±0.13)%,提示细胞凋亡率随药物浓度的增大而增加。CVD10μmol/L组及CVD20μmol/L组与模拟对照组相比,差异有统计学意义。结论 CVD可以抑制HSC-T6的增殖,促进HSC-T6的凋亡。 Objective To investigate the effects of earvedilol (CVD) on HSC-T6 proliferation and apoptosis. Methods HSC-T6 was stimulated by CVD with different concentrations. CCK-8 was used to measured cell proliferation. Cellstaining was done with both Annexin V and PI, The apoptotic rate of different groups was assayed and compared by flow cytometry. Results The proliferation inhibitory rates of different doses of CVD(0 μmol/L,10 μmol/L and 20μmol/L) on HSC-T6 were 2.18 ± 0.60%, 6.30 ± 1.44%, 7.75 ± 1.33%. The difference was statistically significant. The apoptosis rates of different doses of CVD(0μmol/1,10μmol/1 and 20 μmol/1) on HSC-T6 were (2.30 ± 0.08)%, (3.54 ± 0.50)% and (4. 25 ± 0.13)%. The difference was statistically significant. Conclusion Carvedilol concentration-dependent decrease the proliferation and increase the apoptotic rate of HSC-T6 cells.
