应用一次性静脉留置针对大鼠重症急性胰腺炎模型制作方法的改进 被引量:3

摘要 目的探讨一次性静脉留置针对大鼠重症急性胰腺炎(severe acute pancreatitis,SAP)模型制作方法的改进作用。方法 SD大鼠36只,随机分为模型组和假手术组(sham operation,SO),每组18只,采用一次性静脉留置针行胰胆管逆行性穿刺注射5%牛磺胆酸钠制作大鼠SAP模型,分别于造模后3、6和1 2 h处死大鼠,观察两组大鼠死亡率、腹水量;取血清,检测血清淀粉酶变化;取胰腺行组织病理学检查。结果模型组大鼠在各个时间点均出现了不同程度的胰腺组织出血,腺泡细胞坏死,符合SAP的病理变化,造模成功;模型组各个时间点腹水量、胰腺组织病理学评分及血清淀粉酶含量均显著性高于SO组,差异具有统计学意义(P<0.05)。结论一次性静脉留置针可成熟的运用于SAP大鼠模型的制作中,且相对于其他方法具有创伤小、造模时间短、操作简单、不易胰漏等优点,造模结果稳定、重复性好、符合临床特点,具有一定的创新性,值得进一步推广应用。 Objective To investigate related technique improvement on traditional animal model with severe acute pancreatitis(SAP) in rats by disposable intravenous catheter.Methods Thirty-six SD rats were randomly allocated into model group and sham operation group(SO group).5% sodium taurocholate retrograded into bile duct by applying disposable intravenous catheter to setup SAP animal model in rats.Death rate and the volume of ascites were compared between two groups.Serum amylase activity and pathological changes of the pancreas were observed in 3,6 and 12 h postoperatively for both groups.Results The SAP pathological changes were appeared at each time in mode group,modeling successed.The volume of ascites,grade of the pancreatitis histopathology and the concentration of serum amylase in model group were significantly higher than SO group(P 0.05).Conclusion SAP modeling is successfully induced by disposable intravenous catheter.It has the advantage of minimizing the trauma,decreasing the time of modeling,simplifing the procedures.It may be a reliable,long-live model of SAP.
