大七气汤对大鼠子宫内膜异位症P450与COX-2表达的影响

Effect of Da Qiqi decoction on expression of P450 and COX-2 in endometriosis of rats

目的探讨大七气汤加减方(modified da qiqi decoction,MDQD)对子宫内膜异位症大鼠P450、COX-2表达的影响。方法采用SD雌性大鼠子宫内膜自体移植法建模,3周后按随机数字表法分为大七气汤加减方高剂量组(n=9)与低剂量组(n=9)、孕三烯酮组(n=9)、模型组(n=10)及假手术组(n=10)。给药3周后,用发光免疫分析法和ELISA法检测大鼠血清与异位内膜组织雌二醇(E2)与前列腺素E2(PGE2),用免疫组化法和RT-PCR技术检测异位组织芳香化酶P450(P450arom)、环氧化酶-2(COX-2)蛋白与mRNA的表达。结果 MDQD高剂量组大鼠血清E2、PGE2,组织E2、PGE2,P450与COX-2蛋白及mRNA明显低于模型组(P<0.01),与孕三烯酮组比较无明显差异(P>0.05)。结论大七气汤加减方能降低内异症大鼠P450、COX-2和PGE2的含量,通过抑制雌激素生成代谢降低外周血与局部病灶雌激素水平,抑制异位内膜生长。

Objective To investigate the therapeutic mechanism of modified Da Qiqi decoction （MDQD, a traditional herbal prescription）for endometriosis. Methods The endometriosis model was estab- lished in 37 female SD rats and identified with pathological observation. After 3 weeks, they were randomly divided into high dose MDQD group （ n = 9, 5. 072 g/kg）, low dose MDQD group （ n = 9, 1. 268 g/kg）, gestrinone group （n =9, 0.5 mg/kg）, and model group （n = 10）, and another 10 rats with sham-operation served as control. Corresponding agents or normal saline at same volume were given by intragastral administra- tion for 3 weeks. The levels of E2 and PGE2 in the serum and ectopic endometrial tissues were measured by ECLIA and ELISA. The expression of P450 and COX-2 at protein and mRNA levels in the ectopic endometria were determined by immunohistochemistry and RT-PCR. Results Compared with model group, high dose MDQD treatment resulted in significant decline in the E2 and PGE2 levels in the serum and endometrial tissues, and decreased expression of P450 and COX-2 at protein and mRNA levels （ P 〈 0.01 ）. There was no signifi- cant difference in above parameters between the high dose MDQD group and gestrinone group （ P 〉 0.05 ）. Conclusion MDQD reduces the contents of P450, COX-2 and PGE2 in endometriosis rats, which inhibits the growth of ectopic endometria through suppressing estrogenic generation to decrease the level of E2 in serum and local lesions.
