ADAM33基因多态性与尘螨过敏性哮喘易感性关系的研究

摘要 目的探讨人类解整连蛋白和金属蛋白33(adisintegrin and metalloproteinase 33,ADAM33)基因多态性与我国黑龙江省汉族尘螨过敏的支气管哮喘易感性的关系。方法采用引物序列特异性聚合酶链反应-限制性片段长度多态性(PCR-RFLP)的方法检测150例无血缘关系的长期居住在黑龙江省的汉族哮喘患者及120例健康个体ADAM33基因6个位点(T+1,T1,T2,V4,Q-1,S1)单核苷酸多态性。结果在单纯尘螨过敏的哮喘患者组ADAM33基因V4G/C,T2A/G,T1G/A及Q-1A/G(分别是rs2787094 rs2280090 rs2280091 rs612709)等位基因频率高于对照组(P<0.05)。S1和T+1等位基因频率在哮喘患者组和对照组差异无统计学意义。单体型CGGAAG,GGGAAG,CGGGAG,GGGGAG,CG-GAGG,CAGAAG,GGAGAG,CAGAGG,GGG-AGA,GGGGAA,GAAGAG与哮喘相关(P<0.05)。结论ADAM33 V4G/C,T2A/G,T1G/A和Q-1A/G可能参与过敏哮喘的易感性。ADAM33单体型,GGGGAG,GGGAAG,CGGGAG,GGAGAG,GGGAGA,GG-GGAA和GAAGAG可能是哮喘的危险因子,而CGGAGG,CGGAAG,CAGAAG和CAGAGG可能是哮喘的给予保护因子。 Objective To investigate the potential relationship between adisintegrin and metalloproteinase 33(ADAM33) gene polymorphisms and the susceptibility of dust mite allergic asthma happened in Han population of the Heilongjiang province.Methods Six polymorphic sites(V4,T+1,T2,T1,S1,Q-1) were genotyped in 150 non-consanguinity permanent resident patients with asthma in Han population of the Heilongjiang province and 120 healthy individuals as control.Genotypes were determined with the polymerase chain restriction fragment length polymorphism(PCR-RFLP) method.Results The frequency of ADAM33 SNPs T2 A/G,T1 G/A,V4 G/C and Q-1A/G(rs2787094 rs2280090,rs2280091,rs612709) in allergic asthma was higher than those in CTR(P0.05).There was no significant difference in frequency of S1 and T+1 between patients with asthma and CTR.The ADAM33 haplotypes CGGAAG,GGGAAG,CGGGAG,GGGGAG,CGGAGG,CAGAAG,GGAGAG,CAGAGG,GGGAGA,GGGGAA,GAAGAG related with the asthma(P0.05).Conclusion ADAM33 V4 G/C,T2 A/G,T1 G/A and Q-1A/G may participate in the susceptibility of allergic asthma.In addition,the ADAM33 haplo-types,GGGGAG,GGGAAG,CGGGAG,GGAGAG,GGGAGA,GGGGAA,and GAAGAG,may be the risk factors for asthma.However,CGGAGG,CGGAAG,CAGAAG,and CAGAGG may be the protective factors for asthma.
