高效液相色谱法测定六味地黄丸(浓缩丸)中丹皮酚的含量

Study on determination of paeonol content in Liuwei Dihuang pills(condensed) by HPLC

目的建立高效液相色谱(HPLC)法测定六味地黄丸(浓缩丸)中丹皮酚含量的方法。方法采用Dia-monsil TM C18(4.6 mm×150 mm,5μm)色谱柱以十八烷基硅烷键合硅胶为填充剂;甲醇-水(70∶30)为流动相;检测波长为274 nm;流速1.0 ml/mi n;柱温:室温。结果丹皮酚含量在8.24～164.8μg/ml范围内具有良好线性关系,r=0.9999,平均加样回收率为98.92%,相对标准偏差(RSD)=1.70%(n=6)。结论以HPLC法测定六味地黄丸(浓缩丸)中丹皮酚的含量,方法简便快捷,精密度高,重现性好,可用于药品质量的控制。

Objective To establish a HPLC method for determination of paeonol content in Liuwei Dihuang pills （condensed）. Methods The chromatic column of C 18 was applied. The mobile phase consisted of methanol - water （ 70：30） ; the wave length for detection was at 274 nm ; flow rate was kept at 1.0 mL/min. The column temperature kept at room temperature. Results The determination of paeonol content in HPLC had good linear relation in ranges of 8.24 - 164.8 μg/ml （ r = 0. 9999 ） , average recovery rate was 98.92% , and relative standard deviation （RSD） = 1.70% （ n = 6）. Conclusion The results showed that HPLC is convenient, fast, accurate and reproducible in determination of paeonol content in Liuwei Dihuang pills （condensed）. It can be used for controlling the quality of this kind of product of Chinese traditional medicine.
