地塞米松对Lewis肺癌细胞小鼠移植瘤的增殖抑制作用 被引量:1

摘要 目的:探讨地塞米松对Lewis肺癌小鼠肿瘤增殖的影响。方法:将Lewis肺癌细胞接种于C57BL/6小鼠右腋皮下,随机分为3组:对照组、顺铂组和地塞米松组,并给予相应治疗,监测皮下移植瘤体积变化,计算抑瘤率。采用免疫组化法检测肿瘤增殖细胞核抗原(PCNA),细胞周期蛋白D1(Cyclin D1)表达水平;采用流式细胞术(FCM)分析细胞周期变化。结果:各用药组肿瘤的生长明显受到抑制,瘤重明显低于对照组(P<0.05);肿瘤Cyclin D1及PCNA表达水平亦较对照组显著降低(P<0.05);而地塞米松组与顺铂组比较差异无统计学意义(P>0.05)。FCM检测显示,地塞米松使细胞周期明显阻滞于G1期,作用与顺铂相当(P>0.05)。结论:地塞米松对Lewis肺癌小鼠肿瘤增殖具有明显的抑制作用,使细胞在G1期阻滞。 Objective:To observe the effect of dexamethasone on proliferation of Lewis lung carcinoma cells in C57BL/6 mice.Methods:Lewis lung carcinoma cells were inoculated subcutaneously into right axillary of C57BL/6 mice,and the mice were randomly divided into 3 groups:control group,cisplatin group and dexamethasone group.Relevant therapy was given,the change of xenografted tumor volumes was monitored,and the tumor inhibitory rate was calculated.The expression level of proliferating cell nuclear antigen(PCNA) and CyclinD1 in tumor mass were measured by immunohistochemistry.The cell cycle was analysed by flow cytometry(FCM).Results:The tumor growth was suppressed in cisplatin group and dexamethasone group.The weights of tumor were markedly decreased in cisplatin group and dexamethasone compared with control group(P〈0.05).The expression levels of Cyclin D1 and PCNA were significantly lower in cisplatin group and dexamethasone group than control group(P〈0.05).The cells were arrested at G1 phase in dexamethasone group detected by FCM.Meanwhile,there was no significant difference between cisplatin group and dexamethasone group(P〈0.05).Conclusion:Dexamethasone can obviously inhibit the proliferation of Lewis lung carcinoma cells and induce G1 phase arrest.
