摘要 mTOR(mammalian target of rapamycin,mTOR)是雷帕霉素在哺乳动物细胞内作用的蛋白激酶,通过PI3K/Akt信号磷酸化激活而调控细胞分裂、促进转录、信号翻译等。研究表明,mTOR在肿瘤的形成和发展中扮演着非常重要的角色,并参与肿瘤的侵袭和转移。目前,以mTOR为治疗靶点成为肿瘤治疗的研究新热点。本文就近年来有关mTOR靶标及其结合蛋白研究进展作一综述。 The mammalian target of rapamycin(mTOR)has been shown to link growth factor signaling and post-transcriptional control of protein translation through activation of the PI3K/Akt pathway,which is frequently involved in cell cycle progression.mTOR plays a very important role in the formation and development of the tumor and participates tumor invasion and metastasis.Therefore,mTOR-targeted therapy has shown promising role in the management of various cancers.This article reviewed the current status of researches on mTOR and its binding proteins in antitumor therapy.
