老年大鼠组织中Klotho基因表达分布特征 被引量:2

摘要 目的研究Klotho基因在大鼠组织中的表达及其分布。方法饲养SD大鼠分为对照组和老年组,每组6只。处死后取心脏、主动脉、肾脏、脑、肺、肝、胰、脾、骨骼肌、脂肪组织,提取总RNA和总蛋白。应用RT-PCR和ELISA法检测Klotho mRNA及Klotho蛋白在各组织中的表达水平。结果两组大鼠心脏、主动脉、肾脏、脑、肺、肝、胰、脾、骨骼肌、脂肪组织中均有Klotho基因表达,并且在肾脏、胰腺中高表达,脂肪中低表达。老年组Klotho mRNA、Klotho蛋白表达明显减少(P<0.05,P<0.001)。结论大鼠心脏、主动脉、肾脏、脑、肺、肝、胰、脾、骨骼肌、脂肪组织中均有Klotho基因表达。老年大鼠组织中Klotho基因表达减少。 Objective To explore the expression and distribution of Klotho gene in rat. Methods SD rats were randomly divided into control and eider groups. After the model rats were established, heart, aorta, kidney, brain, lung, liver, pancreas, spleen, skeletal muscle, fat were extracted. The expressions of Klotho mRNA and protein were tested by RT-PCR and ELISA. Results Klotho mRNA and protein were expressed in heart, aorta, kidney, brain, lung, liver, pancreas, spleen, skeletal muscle and fat tissue of rat. In comparison with control group, Klotho mRNA and protein expressions in elder group were significantly decreased (P 〈 0. 01 ). Conclusions Klotho mRNA and protein are expressed in heart, aorta, kidney, brain, lung, liver, pancreas, spleen, skeletal muscle and of rat. Expressions of Klotho mRNA and protein in kidney and pancreas are higher,which in fat are lower than those of the other organs. Klotho gene expression in elder are decreased.
