摘要
目的:探讨黄芩素对人子宫颈癌HeLa细胞凋亡的影响及其可能的机制。方法:MTT法测定黄芩素作用后人子宫颈癌HeLa细胞的生长抑制率;Hoechst 33258荧光染色法和Annexin-V/PI双染流式细胞术检测黄芩素诱导HeLa细胞凋亡的作用;Westernblot观察黄芩素对HeLa细胞bcl-2、Bax、caspase 3及p53表达的影响。结果:(1)不同浓度黄芩素作用24h后均可抑制HeLa细胞生长(P<0.05);(2)黄芩素作用24h后,HeLa细胞呈典型的凋亡形态学改变,凋亡率亦显著增加(P<0.01);(3)细胞内Bax、p53和caspase 3表达显著上调,bcl-2表达明显下调(P<0.05)。结论:黄芩素通过调控凋亡相关基因的表达,诱导HeLa细胞凋亡。
Objective:To investigate the effect of baicalein on cell apotosis of human cervix cancer HeLa cells and the possible mechanism.Methods:MTT assay was used to determine the cell growth inhibitory rate.Cellular apoptosis was detected by Hoechst 33258 fluorescent staining and Annexin-V/PI double-staining assay via flow cytometric analysis.The expressions of bcl-2,Bax,p53 and caspase 3 were determined by Western blot.Results:(1)After the treatment with different concentration of baicalein for 24 hours,HeLa cell growth was significantly inhibited(P0.05).(2)The apoptosis rates of HeLa cells were significantly increased in response to baicalein treatment(P0.01).(3)The expressions of Bax,p53 and caspase 3 were up-regulated(P0.05 to P0.01),but the expression of bcl-2 was down-regulated when HeLa cells were treated with baicalein for 24 hours.Conclusion:Baicalein can induce apoptosis of HeLa cells probably by regulating the expressions of gene assioated with apoptosis.
