As_2O_3联合热疗对人食管癌EC-1细胞增殖和细胞周期的影响

摘要 目的:探讨As2O3联合热疗对人食管癌细胞株EC-1增殖的抑制作用及机制。方法:人食管癌细胞株EC-1细胞分为对照组、As2O3组、热疗组及As2O3+热疗组。采用MTT法测定细胞的增殖情况,应用流式细胞术检测细胞周期变化,TUNEL法观察细胞凋亡。结果:与对照组相比,As2O3组和(或)热疗组24、48、72h后的EC-1细胞生存率均降低,As2O3+热疗组的细胞生存率低于单纯As2O3组和热疗组(P<0.05或P<0.01)。As2O3+热疗组G0/G1期细胞低于热疗组,G2/M期细胞比例高于热疗组和As2O3组,As2O3+热疗组S期细胞比例低于As2O3组,差异均有统计学意义(P<0.05或P<0.01)。As2O3组和As2O3+热疗组的细胞凋亡率均高于对照组,As2O3+热疗组高于As2O3组和热疗组,差异均有统计学意义(P<0.05或P<0.01)。结论:As2O3联合热疗具有协同抗食管癌作用,其机制可能与联合应用后增强诱导食管癌细胞凋亡、细胞周期特异性治疗及细胞周期调控剂联合增效有关。 Objective: To study the effect of As2O3 combined with hyperthermia on the growth of esophageal carcinoma EC-1 cell line. Methods: EC-1 cells were divided into four groups: control group, As2O3 group; hyperthermia group and As2O3 combined with hyperthermia group. The cell proliferation was detected by MTT assay. The cell cycle and the apoptosis were observed by flow cytometry and TUNEL assay. Results: Compared with control group,cell survival rates of EC-1 ceils were decreased after 24, 48 and 72 h treatment in As2O3 group and hyperthermia group. The cell survival rate was lower in As2O3 combined with hyperthermia group than that of As2O3 group and hyperthermia group (P 〈 0.05 or P 〈 0.01). The ratio of ceils in G0/G1 phase was lower in As2O3 combined with hyperthermia group than that of hyperthermia group. The ratio of ceils in G2/M was higher in As2O3 combined with hyperthermia group than that of As2O3 group and hyperthermia group. The ratio of ceils in S phase was also significantly lower in As2O3 combined with hyperthermia group than that of As2O3 group (P 〈 0.05 or P 〈 0.01). The apoptotic rates were higher in As2O3 group and As2O3 combined with hyperthermia group than that of control group. The apoptotic rate was significantly higher in As2O3 eombined with hyperthermia group than that of As2O3 group and hyperthermia group (P 〈0.05 or P 〈 0.01). Conclusion: As2O3 in combination with hyperthermia can produce synergistic killing effect on EC-1 cells, which may be related with the increase of apoptosis, Cell cycle specific killing and cell cycle regulator.
