摘要
目的:用蛋白质组学的方法研究壮骨止痛方治疗骨质疏松的作用机理。方法:建立去卵巢大鼠骨质疏松症模型,分为壮骨止痛方组、模型组、假手术组,13周后骨质疏松症模型成功,提取股骨组织蛋白质样品,双向电泳分离,得到各组股骨组织总蛋白质分子解剖图谱,用图像分析软件,分析各组间差异蛋白质点,MAlDI-TOF-MS质谱分析,结合蛋白质生物信息库(Marx science Ltd database),对各蛋白质初步鉴定。结论:鉴定了2个差异蛋白,分别P1为膜联蛋白A1,P2为蛋白质二硫键异构酶A3,初步认为这两种蛋白质在壮骨止痛方治疗该病过程中发挥着重要调控作用。
Objective:To study the mechanism about Zhuanggu Zhitong Formula treating OP based on proteomics. Methods : Bilateal OVX in rats was performed as osteoporosis model. Rats were randomly divided into three groups : control group, OP model group and Zhuanggu Zhitong Formula group. The pathology of bone was examined after 13 weeks. In the present proteomie study, we characterized the protential effects of OVX and Chinese medicine on protein expression in ratbones. Using two dimensional electrophoresis,mass spectrometry and Marx science Ltd database, we elementarily identi- fied three variational proteins. Conclusions:Two proteins were identified as proteins similar to Annexins A1, Protein disul- fide- isomerase A3. These proteins have been demonstrated to be postmenopausal proteins. These results can provide valuable experimental evidences for the research of the molecular mechanism of Zhuanggu Zhitong Formula treating OP.
