摘要
目的观察印度刺猬蛋白(Ihh)对成骨细胞p38丝裂素活化蛋白激酶(p38MAPK)的调节作用。方法取子鼠颅盖骨分离培养成骨细胞,采用实时定量聚合酶链反应(Real-timePCR)检测成骨细胞中p38MAPK的表达量,检测成骨细胞在不同浓度(0、10、50、200μg/L)及不同时问(0、1、2、3d)的Ihh作用下p38MAPK的表达,采用噻唑蓝(MTT)法检测细胞增殖。结果成骨细胞表达p38MAPK;Ihh促进成骨细胞增殖,促进p38MAPK的表达(P〈0.05);阻断p38MAPK信号转导通路后,成骨细胞增殖明显受抑制,但不能明显抑制Ihh刺激下的成骨细胞增殖(P〈0.05)。结论p38MAPK在成骨细胞增殖分化中起作用,Ihh通过p38MAPK通路促进成骨细胞增殖。
Objective To explore the effect of Indian hedgehog (lhh) on the expression of p38 mitogen-activated protein kinase ( p38 MAPK) in osteoblasts. Methods The calvarial of neonatal rats was sampled, and the osteoblasts were cultured. The expression levels of p38 MAPK in the osteob]asts treated with differen: concentrations (0, 10, 50 and 200 μg/g/L) of Ihh or various durations (0, 1,2 and 3 days) were detected by using real-time polymerase chain reaction. The proliferation of osteoblasts was quantified by using methyl thiazol tetrazolium (MTI') assay. Results p38 MAPK was expressed in the osteoblasts. Ihh increased p38 MAPK mRNA expression level in osteoblasts in vitro, p38 MAPK inhibitor could inhibit the proliferation of osteoblasts, but eouldn' t suppress the Ihh-induced prolifation of osteoblasts. Conclu- sion The proliferation and differentiation of osteoblasts were promoted by p38 MAPK. Ihh can enhance the proliferation of osteoblasts via p38 MAPK pathway.
