Modified technique for abdominal heart transplantation in rats

摘要 目的改良大鼠腹腔心脏移植术式,建立符合生理条件、稳定的动物模型。方法取封闭群雄性SD大鼠248只为供受体,进行同种同基因腹腔心脏移植术,主肺动脉、左肾静脉的吻合方法为改良袖套法。结果定向实验阶段共完成84例大鼠腹腔心脏移植手术,供心热缺血时间0min,冷缺血时间(26.63±1.14)min,并发症主要为术中大出血、血栓形成。手术成功率为88.7%(72/84),72例术后7d存活率为100%。结论改良大鼠心脏移植手术能明显缩短供心冷缺血时间,保证吻合口无出血,从而提高手术成功率。 Objective To modify the technique for abdominal heart transplantation (AHT) in rats and establish a physiological and stable animal model. Methods A total of 248 SD rats served as donors and recipients to establish homogeneous and heterotopic models of AHT. Anastomosis of main pulmonary artery and renal vein was done by using the modified cuff technique. Results In 84 AHT models of AHT in rats, the average warm ischemic time was 0 min, and the average cold ischemic time was (26. 63 ± 1.14) min. The operation was successfully carried out in 72 rats and failed in 12 rats with the successful rate being 88.7 % (72/84). The most frequent complications were bleeding in the operation, and thrombo- sis in aorta and renal vein after transplantation. The survival rate ( 〉7 days) was 100%. Conclusion Modified technique for AHT in rats reported herein simplified the procedure and shortened cold ischemic time, decreased the loss of blood during operation and increased the successful rate.
