CDKN2A基因启动子区甲基化对乳腺癌的影响及其临床意义

摘要 目的:研究细胞周期依赖性激酶抑制基因(CDKN2A)启动子区甲基化对乳腺癌的影响及其临床意义。方法:应用甲基化特异性聚合酶链反应(PCR)、West ern-bl ot检测乳腺癌及癌旁组织65例,比较CDKN2A基因启动子区甲基化及其蛋白质与乳腺癌发生发展的关系。结果:乳腺癌组织CDKN2A甲基化产物明显多于癌旁组织,差异有统计学意义(χ2=13.7,P=0.001);乳腺癌组织和癌旁组织CDKN2A mRNA甲基化产物的PMR水平差异有统计学意义(t=3.37,P=0.001);乳腺癌组织CDKN2A蛋白的表达水平显著低于癌旁组织(t=2.784,P=0.012)。结论:CDKN2A基因启动子区甲基化及其编码蛋白质的失活与乳腺癌明显相关。 Objective To analysis clinical implications of methylation of the cell cycle dependence kinase suppressor genes (CDKN2A) on promoter area in patients with breast cancer. Methods The organizations of 65 breast cancer and next to the organization of the cancer were studied. The expression of methylation of CDKN2A gene on promoter area was detected by polymerase chain reaction (PCR).The protein expression of CDKN2A was determined by western blot analysis. Results The product of methylation of CDKN2A gene on promoter area of the breast cancer organizations was significantly higher than the next organization (the chi-square=13.7,P=0.001);the PMR level of CDKN2A gene in patients with breast cancer between the breast cancer and next to the organization of the cancer was statistically significant (t=3.37, P=0.001);the CDKN2A protein expression level of breast cancer organizations was significantly less than next to the organization of the cancer (t=2.784,P=0.012). Conclusion The methylation of CDKN2A gene on promoter area and the inactivation of CDKN2A protein are significantly related to breast cancer.
