糖尿病大鼠甲状腺组织核因子-κB、Bax、Bcl-2表达的变化

摘要 目的通过测定糖尿病(DM)大鼠核因子-κB(NF-κB)、Bax、Bcl-2表达的变化来探讨甲状腺功能损伤的可能机制。方法 14只雄性Wistar大鼠随机分为糖尿病组(DM组)8只和对照组(N组)6只。DM组大鼠成模后12周末杀检。免疫组织化学方法观察两组大鼠甲状腺组织中NF-κB、Bax、Bcl-2表达的变化。结果光镜下N组甲状腺滤泡上皮细胞核NF-κB染色为阴性,而DM组细胞核内染色呈阴性或弱阳性,图像分析结果,DM组甲状腺组织NF-κB染色的平均光密度和阳性面积比均较N组升高(P<0.05),DM组Bax染色阳性面积比较N组增加,两组间Bax染色平均光密度比较差异无统计学意义。DM组Bcl-2免疫组织化学染色阳性面积比和平均光密度均较N组下降(P<0.05)。DM组Bcl-2/Bax的阳性面积比和平均光密度比均较N组下降(P<0.001)。结论 DM状态下NF-κB异常激活以及甲状腺滤泡上皮细胞凋亡增多是甲状腺损伤的可能机制之一。 【Objectives】 By detecting the expression changes of nuclear factor-κB,Bax,Bcl-2 in diabetic rat thyroids to investigate the possible mechanism of the diabetic rat thyroid function impairment.【Methods】 14 male Wistar rats were divided randomly into DM group(n =8) and N group(n =6).12 weeks later after administration,rats were sacrificed,then detected the expression Changes of NF-κB,Bax,Bcl-2 by immunohistochemistry.【Results】 under light microscope,NF-κB was week expressed in the nucleus of thyroid follicular epithelial cell of DM group.And in the normal group,NF-κB was negatively expressed in the nucleus,the results of the image analysis: the ratio of the positive reactive area and the average optical density of NF-κB in DM group were both increased(P 0.05) as compared with the N control group.There isn't significant deviation between the average optical densities of Bax in the two groups.The ratio of the positive reactive area of Bax in DM group were increased(P 0.05).The ratio of the positive reactive area and the average optical density of Bcl-2 in DM group decreased(P 0.05) as compared with N group.Compared to the N group,the ratio of Bcl-2 and Bax in DM group decreased(P 0.001).【Conclusion】 It is possible that the unusually activated NF-κB and apoptosis participated the mechanism of thyroid function impaired in diabetes mullitus group.
