响应面法优化马血中SOD热变性提纯工艺研究

Optimization of heating denaturation extraction technique of SOD from horse blood by using response surface methodology

摘要 以SOD比活力为响应值,在单因素实验的基础上,筛选出热变温度、热变时间、Cu2+添加量3个对提纯影响较大的因素,响应面法进行优化,并验证优化方案。结果表明,SOD热变性提纯的优化参数为:通过实验,热变温度为55℃,时间为15.5min,Cu2+添加量为2.08%。在此条件下,SOD比活力为700.58U/mg。 Based on single factor experiment, the report screened out three main factors, including the heating denatureation temperature, heating denatureation time, and amount volume of Cu^2+ at first optimization of these factors by using response surface methodology. Results of response surface analysis showed that the optimum heating denatureation extraction conditions of SOD were as follows: heating denatureation temperature 55℃, heating denatureation time 15.5 rain, amount volume of Cu2+ 2.08%. Through this condition, specific activitv of SOD to 700.58 U/ma.
