Determination of Cordycepin in Cultured Cordyceps Medium by HPLC

摘要 建立蛹虫草培养基中虫草素的含量测定方法。采用反相高效液相色谱法,以十八烷基键合相硅胶为固定相,0.01mol/L的磷酸二氢钾溶液(pH=6.0)-乙腈(93:7,V/V)为流动相,检测波长260nm,流速1.0mL/min进行分析。结果虫草素在2.0—40.0μg·mL-1范围内线性良好(r=0.9999);平均加标回收率分别为96.8%、98.1%和100.9%,RSD分别为3.33%,2.88%和1.60%(n=3);方法精密度RSD为0.29%(n=5)。该法分析时间短,结果准确,重现性好,适合人工蛹虫草培养基中的虫草素的含量测定。 The method for determination of the content of cordycepin in cultured cordyceps medium was established. The compound was determined by reversed-phase high performance liquid chromatography with octadecyl bonded silica as stationary phase and mobile phase of phosphate buffer solution (0.01mol · L-1 ,pH=6.0)-acetonitrile (93 : 7,V/V) at a flow rate of 1.0mL · min-1 ,and detection wavelength was 260 nm. The cordycepin had the good linearity in the range of 2.0--40.0μg 100.9%with (n = 5). This determination · mL- 1 (r = 0.9999). The recoveries of cordycepin were respectively 96.8 %, 98.1% and RSDs of 3. 33% ,2.88%and 1.60%(n=3) ,and the precision of the method was 0.29% method has rapid analysis, accurate results and good repeatbbility, and is suitable to of the content of cordycepin in the cultured cordyceps medium.
