摘要
利用光谱法对4-羟基香豆素(4-HC)和杀鼠剂溴敌隆(BRD)、溴鼠灵(BDF)与牛血清白蛋白(BSA)的相互作用进行了研究。依据Foerster能量转移理论计算了3种药物与BSA212位Trp残基的结合距离。利用光谱探针,通过竞争实验判断4HC、BRD、BDF主要结合于BSA的Subdomain IIA结构亚域的Site I。实验初步探讨了该类药物与蛋白的结合模式与作用机制。
The interactions between bovine [4-hydroxylcoumarin (4-HC), bromadiolone (BRD) serum albumin (BSA) and three drugs and brodifacoum (BDF)] were studied by spectroscopy. The binding distances between BRD, BDF, 4-HC and Trp-212 residue on BSA were theoretically calculated based on theory of Foerster energy transfer. Three drugs binding location on BSA was confirmed by competition test, drugs known binding Sites were obtained by fluorescence probe. It revealed that BRD,BDF and 4-HC had mainly bind on Site I in Subdomain II A of BSA. In addition,the binding mode and interaction mechanism of the drugs with BSA were discussed.
