双糖链蛋白聚糖对大肠癌细胞生长作用研究

摘要 目的研究Biglycan对大肠癌细胞HT-29和SW480侵袭、凋亡等生长作用影响。方法采用Transwell方法检测Biglycan对大肠癌细胞HT-29和SW480的侵袭能力影响;AV/PI染色检测细胞凋亡情况;Western blot法检测加药处理前后细胞内Rb、pRb和Ras表达变化。结果 100 nmol/L Biglycan和50 nmol/L Biglycan可抑制大肠癌细胞HT-29和SW480侵袭转移,同时可诱导HT-29、SW480细胞发生凋亡;经Biglycan处理后,细胞内Rb总蛋白无显著性变化,pRb和Ras出现下调趋势。结论 Biglycan可抑制大肠癌细胞HT-29和SW480侵袭转移、诱导细胞凋亡,下调pRb和Ras的表达、调控细胞周期从而抑制细胞生长为其可能的作用机制。 Objective To observe the effect of biglycan on the invasion and apoptosis of HT- 29 and SW480 cells in vitro. Methods Transwell assay was used to detect the cell invasion. The ap- optosis was determined by AV/PI staining, the cell cycle was determined by PI staining and the ex- pressions of Rb, pRb and Ras were detected by Western blot. Results The results showed that 100 nmol/L and 50 nmol/L Biglycan could induce inhibition of invasion and apoptosis in the HT-29 and SW480 cells, respectively. Compared with the control group, the expression of Rb showed no signifi- cant difference, and the expressions of pRb and Ras were decreased. Conclusion Biglycan inhibits colon cancer cells invasion, induces apoptosis and might be related to down-regulated expressions of pRb and Ras and regulation of cell cycle.
