摘要
目的探讨辅助性T细胞(Th)9相关细胞因子在类风湿性关节炎(RA)中表达的改变及可能机制。方法采集15例健康人(健康对照组)和15例RA患者外周血,以酶联免疫吸附法测定血清白细胞介素(IL)-9、IL-4、转化生长因子-β(TGF-β)和干扰素-γ(IFN-γ)水平,分析这些因子在Th9分化中的作用。结果 RA患者血清IL-9水平与健康对照组比较差异无统计学意义(P>0.05);RA患者血清TGF-β水平较健康对照组显著减低,差异有统计学意义(P<0.01);RA患者血清IL-4水平较健康对照组有所降低,但差异无统计学意义(P>0.05);IFN-γ水平在5例RA患者外周血中可以检出,其余低于可检出浓度,健康对照组均低于可检出浓度。结论 RA患者外周血Th9细胞的促分化因子降低,抑分化因子升高,导致Th9细胞缺乏分化的微环境,由此推测循环血Th9细胞不参与RA的发病环节。
Objective To observed the change and possible function of helper T cell 9 (Th9) cells in the pathogenesis of rheumatoid arthritis( RA ). Methods Serum samples were collected from 15 RA patients and 15 healthy controls. The levels of interleukin(IL)-9, IL-4, transforming growth factor(TGF)-β and interferon(IFN)-γ in the subjects were tested by enzyme- linked immunosorbent assay. Results There was no statistically significant difference in the levels of IL-9 and IL-4 between the two groups(P 〉 0.05). The level of TGF-β in RA patients was significantly lower than that in healthy control group( P 〈 0. 01 ). IFN-γ was detectable in five cases of RA, while other samples of RA and healthy controls were not detectable. Conclu- sion Differentiation factors reducing and inhibiting differentiation factors increasing can result in the lack of suitable microen- vironment for polarization of Th9 cells in peripheral blood of RA patients, which imply that circulating Th9 cells may not be in- volved in the pathogenesis of RA.
