摘要
目的:探讨苦参碱诱导宫颈癌Hela细胞凋亡的机制。方法:将Hela细胞接种于6孔培养板内的盖玻片上,以1.0、1.5、2.0mg/ml Mat作用于Hela细胞72h后,用免疫细胞化学法(SP法)检测药物作用后细胞中Bcl-2、COX-2、Fas、FasL蛋白的表达水平。结果:以1.0、1.5、2.0mg/ml Mat作用Hela细胞72h后,Bcl-2、COX-2、FasL蛋白表达随着Mat作用剂量增加而逐渐下降,Fas蛋白表达随着Mat作用剂量增加而增强;蛋白阳性表达率与对照组相比,均差异有统计学意义。结论:苦参碱诱导宫颈癌Hela细胞凋亡可能与下调COX-2,Bcl-2和FasL表达,上调Fas表达有关。
Objective:To explore the probable mechanisms of matrine in induction of cervical carcinoma Hela cells apoptosis.Methods: Hela cells were planted glass slices,after being treated with(1.0、1.5、2.0mg/ml) matrine,immune-ohistochemistry method was performed to determine the expression of Bcl-2,COX-2,Fas,FasL proteins.Results: Immunohistochemitry assay discovered,after being treated with 1.0、1.5、2.0mg/ml matrine for 72h,the expression of Bcl-2、COX-2 and FasL protein were down-regulated,while that of Fas protein was up-regulated in Hela cells,with concentration of matrine increasing.Compared with control group respectively,all positive expressions for Bcl-2,COX-2,Fas and FasL had significant difference.Conclusions: Matrine can significantly inhibit proliferation of cervical carcinoma Hela cells,and induce apoptosis of them.The mechanisms may be as follow: by down-regulating the expression level of Bcl-2、COX-2 and FasL,up-regulating that of Fas apoptosis related protein.
