摘要
为了探讨猪第二极体是否具有支持胚胎发育的全能性,采用显微操作的方法将猪第二极体移入去除雌原核的原核胚中,电融合完成重组,体外观察重组胚胎的发育情况。结果显示:猪卵母细胞体外受精4 h后即有第二极体排出;受精20 h后,50.6%的受精卵排出第二极体,且82.2%的第二极体具有活性。将有活性的第二极体与去除雌原核的原核胚重组后,重组胚胎能够发育到8-细胞期,并且重组胚胎发育的8-细胞与体处受精胚胎发育的8-细胞在形态上没有明显差别。表明猪第二极体核物质能够支持胚胎发育。
To discuss whether porcine second polar body(Pb Ⅱ) could support embryonic development or not,the second polar bodies were injected into pronuclear embryos without female pronucleus by micromanipulation,and Pb Ⅱs and pronuclear embryos were fused by electrofusion.The recombinant embryos were cultured in vitro and their ability of further development were observed.The results showed that porcine oocytes extruded Pb Ⅱs at 4 h after in vitro fertilization,and 50.6% of fertilized oocytes exhausted Pb Ⅱs at 20 h,82.2% Pb Ⅱs being active.The recombinant embryos,which were active Pb Ⅱs being injected into pronuclear embryos without female pronucleus,could develop to 8-cell stage.The 8-cell embryos coming from recombinant embryos had the same feature with in vitro fertilized 8-cell embryos.In conclusion,the nuclear materials of porcine second polar body could support the early development of embryos.
