鸭肉肌内磷脂酶解反应条件优化及其产物鉴定 被引量:2

摘要 以磷脂脂质体为反应介质及鸭肉肌内磷脂酶为酶液进行考察磷脂酶解的影响因素。以脂肪酸条带分析的Trace Qty值为指标,通过正交试验,筛选出鸭肉肌内磷脂酶解的最佳反应条件。结果表明,磷脂酶解的最佳反应条件为酶与底物比例4∶1、温度45℃、超声频率45 KHz、超声功率270 W、反应时间0.5 h。液相色谱质谱联用仪(LC-MS)在磷脂酶解产物中检测到油酸的生成,进一步证实鸭肉内源磷脂酶的存在。 The factors affecting enzymatic hydrolysis of intramuscular phospholipase were determined with the reaction system consisting of intramuscular phospholipase extracted from duck muscle and liposome substrate.The orthogonal experiments were carried out and the hydrolysates were identified with Tract Qty to identify the optimal reaction condition.The phospholipid hydrolysis reached the optimum at the enzyme and substrate ratio of 4∶1,temperature of 45 ℃,ultrasonic frequency of 45 KHz,ultrasonic power of 270 W,and reaction time of 0.5 h.Oleic acid was present in the hydrolysates as detected by liquial chromatography-mass spectrometry(LC-MS),which further confirmed the existence of endogenous phospholipase system.
