刀豆蛋白A对帕金森患者外周血单个核细胞分泌IL-10和IFN-γ的作用

Effect of Concanavalin on IL-10 and IFN-γ Secretion of Peripheral Blood Mononuclear Cells in Patients with Parkinson's Disease

摘要 目的:观察帕金森病(PD)病人外周血单个核细胞(PBMC)经刀豆蛋白A(Co-nA)诱导培养后分泌白介素-10(IL-10)和干扰素-γ(IFN-γ)的能力。方法:23例PD患者分为早中期组(12例)和晚期组(11例),24例健康人群作为对照组,分离各组外周血单个核细胞,并用ConA诱导刺激。采用ELISA分别检测各组培养上清液中IL-10和IFN-γ含量。结果:对照组和早中期组PD患者经ConA诱导培养的PBMC分泌IL-10水平显著高于普通培养(P<0.05);晚期组分泌IL-10水平明显低于对照组和早中期组(P<0.01)。各组PD患者PBMC分泌IFN-γ水平ConA培养比普通培养都有升高,早中期组最高(P<0.01);PD早中期组ConA培养的PBMC分泌IFN-γ水平显著高于对照组和晚期组(P<0.01)。结论:刀豆蛋白对PD患者PBMC分泌IL-10和IFN-γ有促进作用;免疫机制在PD发病和发展中起着重要的作用。 Objective: To investigate the capability of peripheral blood mononuclear cells (PBMC) to produce IL-10 and IFN-γ induced by concanavalin a(ConA) in Parkinson disease(PD) patients. Method: 23 PD patients were divided into early or mid-stage group (n=12) and the late stage group (n=11),24 healthy people as a control. ELISA was adopted to detect the IL-10 and IFN-γ content in each group. Results: IL-10 secretion from PBMC in PD patients which on the early and mid-stage group when stimulated with ConA culture was significantly higher than in the presence of pure RPMI-1640 culture (P<0.05). There were significantly differences among IL-10 secretion levels of the late stage group compared with that of the healthy control group and early and mid-stage group(P<0.05). All the groups of PD patients PBMC secrete IFN-γ were increased, which the highest concentration of IFN-γ in the early and mid-stage group in the presence of ConA culture conditions, significantly higher than in the presence of pure RPMI-1640 (P<0.01). The PD in the early and mid-stage group PBMC secreting IFN-γ , compared with the control group, was significantly increased(P<0.01) . Conclusion: ConA can stimulate IL-10 and IFN-γ secretion in PBMC of PD patients. Abnomal immune function was one of the mechanisms involved in the pathogenesis and development of PD.
