摘要
目的:观察内皮源性吲哚胺2,3-过氧化酶(IDO)对自体移植静脉桥狭窄的抑制作用。方法:成年长耳大白兔32只,随机分为实验组和对照组,每组16只。实验组利用建立内皮细胞高表达IDO的颈静脉作为血管桥,对照组则利用质粒载体建立静脉桥。分离并切取2cm长颈外静脉,移植到自体同侧颈总动脉中。分别于术后当天、第7天、第14天和第21天取两组动物各4只,完整取下移植静脉桥,常规石蜡包埋,切片后分别行HE染色及PCNA免疫组化染色,计算静脉桥中膜厚度和平滑肌细胞PCNA阳性指数。结果:两组移植静脉桥中膜均有不同程度增厚,且有随移植时间延长而增加的趋势。移植后第21天实验组静脉桥中膜厚度测值明显小于对照组(P<0.05),且平滑肌细胞PCNA阳性指数明显低于对照组(P<0.05)。结论:内皮源性IDO对移植静脉内平滑肌细胞增生有抑制作用。
Objective: To discuss that the indoleamine-2,3-dioxygenase inhibits autogeneic vein graft model. Method: 32 adult rabbits used in this study were divided into two groups (n= 16). A single administration of SB-hIDO complex was given before the vein graft model were set up in rabbits. The vehicle was givens as the controlled group. Of all rabbits the autogenous jugular vein was grafted into the carotid artery by microsurgery. After the operation each group were fed in the same way. At 7 d, 14 d and 21d after grafting, from four of each groups the grafted vessel were taken out. Results: Intimal hyperplasia (IH) of vein graft in experiment group increaed faster than that in control group. And the anti-proliferating cell nuclear antibody (PCNA) index of smooth muscle cell (SMC) in IH of experiment group was also lower than that of contr ol group. Conclusion: Enhanced endothelial IDO can ameliorate the stricture of the vein graft model.
