摘要
目的观察龟鹿二仙胶及其拆方对豚鼠膝关节软骨功能基因表达的影响,探讨龟鹿二仙胶的配伍机理。方法取3月龄豚鼠膝关节软骨建立软骨细胞体外培养体系,采用免疫组化染色、RT-PCR、Elisa方法检测酸性GAG、Ⅱ型胶原、Bax、Bcl-2、iNOs的表达。结果龟鹿参组能显著促进软骨细胞分泌酸性GAG(P<0.05),促进Ⅱ型胶原合成(P<0.05);龟鹿杞组能显著抑制Bax的表达、促进Bcl-2表达,抑制软骨细胞分泌iNOs(P<0.05),二组的作用与龟鹿二仙胶组相同。结论人参可助龟甲胶、鹿角胶促进软骨细胞分泌酸性GAG和Ⅱ型胶原,枸杞子可助龟甲胶、鹿角胶延缓软骨细胞凋亡,抑制软骨细胞分泌iNOs。
Objective To observe the effects of Guilu Erxian glue and its separated recipes on gene expression in chondrocytes of knee joint of cavia cobaya,and explore its compatibility mechanism.Methods The knee joint cartilage of 3 month old cavia cobaya was used to establish the culture system of chondrocytes in vitro.The expressions of acidic GAG,collagen type Ⅱ,Bax,Bcl-2 and iNOs were detected by immunohistochemical staining,RT-PCR and Elisa.Results The expressions of acidic GAG and collagen type Ⅱ were increased in Guilushen group(P0.05).The expressions of Bax and iNOs were decreased,and the expression of Bcl-2 were increased in Guiluqi group(P 0.05).The effects of the two groups were same to that of Guilu Erxian glue group.Conclusion Colla carapacis et plastri testudinis and colla cornus cervi can promote chondrocytes to secrete acidic GAG and collagen type Ⅱ,and ginseng is helpful for the effects.Colla carapacis etplastri testudinis and colla cornus cervi can delay the apoptosis of chondrocytes,inhibit chondro cytes to secrete iNOs,and lycum barbarum is helpful for the effects.
