摘要
目的筛选雷公藤叶提取物中具有抗肿瘤活性的部位。方法以乙醇为溶剂,超声提取雷公藤叶,所得粗提物以大孔树脂分离纯化;用噻唑蓝比色法(MTT)评价纯化后各洗脱部位对人乳腺癌细胞(MCF-7)和前列腺癌细胞(PC3)增殖的抑制作用。结果雷公藤叶提取物70%乙醇洗脱部位(TAE70)在体外对MCF-7、PC3细胞的最大抑制率为61.96%、67.28%;雷公藤叶提取物95%乙醇洗脱部位(TAE95)对MCF-7、PC3细胞的最大抑制率为84.09%、88.77%。结论 TAE70、TAE95是2个体外抗肿瘤活性较强的组分。
Objective To screen the anti-tumor active components in extract of Tripterygium wilfordii Hook.f.leaf.Methods Samples were extracted with alcohol by ultrasound,and the crude extraction was purified by macroporous resin.The inhibitory effects of elution parts on proliferation of MCF-7 and PC-3 cells were evaluated by MTT assay.Results For MCF-7 and PC-3 cells,the maximal inhibition rate of the proliferation by 70% ethanol elution part(TAE70) was 61.96% and 67.28%,and the maximal inhibition rate of the proliferation by 95% ethanol elution part(TAE95) was 84.09% and 88.77% in vitro.Conclusion TAE70 and TAE95 were choosed as the components which have strong anti-tumor activity in vitro.
