金针菇双单杂交的遗传分析 被引量:7

摘要 以3个金针菇为材料,研究其单核原生质体不亲和因子基因型,双单杂交构建杂交种,并对其基因型和遗传差异进行分析。结果表明,3个金针菇菌株的单核原生质体形成率不同,单核原生质体率分别为31.9%、88.5%和13.4%;不亲和因子基因型为2种类型。以F32和F21的单核原生质体配对构建双核体,再与F2121的单核原生质体菌株配对构建杂交种,杂交亲和率为100%;双单杂交种与亲本双核体杂交存在拮抗反应,而杂交种杂交彼此无拮抗反应。双单杂交种的单核原生质体不亲和因子基因型分析显示,杂交种的基因型一个与受体相同,另一个与供体的一个基因型相同。ISSR分析显示,双单杂交种与其单核原生质体和具有相同基因型的单单杂交种亲缘关系较近。 Three Flammulina velutipes stains were used for studying the genotype of the incompatibility factors of the mononuclear protoplasts and di-monad breeding,the differences on genotype and genetic diversity of the three Flammulina velutipes strains were also analyzed.The result showed that the protoplast formation rate of three strains were 31.9 %,88.5 % and 13.4 %,respectively.Among their genotypes,there were two genotypes of the incompatibility factors.The hybrid species were constructed by mating of the mononuclear protoplast F2121 with dikaryotic strain which was bred through F32 and F21,and the compatibility rate of combination was up to 100 %.The antagonistic reaction existed between di-monad hybrids and the dikaryotic,but did not exist among di-monad crossing hybrids.The genotype analysis of incompatibility factor in mononuclear protoplast of di-monad hybrids showed that one genotype of the hybrid species was the same as the receptor,and another was the same as donator.ISSR(inter-simple sequence repeat) analysis indicated that the genetic relationship between di-monad crossing hybrids or its mononuclear protoplast and the same genotype monosporous hybrids was close.
