γ-氨基丁酸A受体及受体基因在大鼠急性脑缺血中的变化 被引量:2

Changes of the expression of GABAAR and GABAARα1 mRNA induced by acute cerebral ischemia in rats

摘要 目的观察大鼠大脑皮质γ-氨基丁酸A受体(GABAAR)及受体基因(GABAARα1mRNA)在脑缺血不同时段的表达及其动态变化的规律,探讨其在急性缺血性脑血管病中变化的意义。方法 45只雄性Wistar大鼠随机分为对照组,脑缺血1h组、6h组、24h组、3d组(均为手术组),假手术组;采用改良的大鼠大脑中动脉栓塞(MCAO)模型,至规定时间点取大鼠脑组织,分析各组GABAAR及GABAARα1mRNA在大脑皮质阳性神经元的表达情况。结果各手术组GABAAR及GABAARα1mRNA表达均低于假手术组及对照组,与对照组相比,GABAAR在缺血1h其表达既有降低,至6h降至最低。GABAARα1mRNA表达在缺血24h开始下降,3d下降更明显,而假手术组与对照组相比差异无统计学意义。结论 GABAAR及GABAARα1mRNA在急性缺血时含量降低可能为缺血性脑血管病的重要原因。 Objective To investigate the change of GABAAR and GABAARα1 mRNA and the dynamic regulation in the cortex of rats with focal cerebral ischemia in different phases and to observe its means in acute ischemia.Methods Forty-five male rats were randomly divided into three groups: the normal control,sham operation and ischemic operation group(subdivided into 1 hour,6 hours,24 hours and 3 days groups) by middle cerebral artery occlusion model.Brain specimens of the rats from each group were obtained to observe the damage of neurons and to study the positive expression of neurons,and the positive cell number expressed by GABAAR in each group and the changes of GABAARα1 mRNA expression were observed.Results Compared with the normal control group and sham operation group,the expressions of GABAAR and GABAARα1 mRNA in each ischemia operation group were lower.The expression of GABAAR was decreased after 1 hour and reached the lowest at 6 hours after ischemia.The expression of GABAARα1 mRNA was decreased from 24 hours and was the bottom within 3 days.However,there was no difference between the normal control group and sham operation group.Conclusion It is suggested that the decreasing of the contents of GABAAR and GABAARα1 mRNA after ischemia might be related with ischemia injury.
