Effects of DEHP and MEHP on apoptosis of leydig cells of mice

摘要 目的探讨邻苯二甲酸二(2-乙基)己酯(DEHP)及其代谢产物邻苯二甲酸单(2-乙基)己酯(MEHP)对小鼠睾丸间质细胞凋亡的影响。方法用不同浓度的DEHP(0、1、10、100μmol/L)及MEHP(0、1、10、100μmol/L)分别处理小鼠睾丸间质细胞48h,应用流式细胞仪AnnexinV FITC-PI双染法分析各组细胞凋亡情况。结果100μmol/L DEHP组及10,100μmol/L MEHP处理组与0μmol/L组比较,凋亡细胞比例明显增加(P<0.05);活细胞比例明显减少(P<0.05);10、100μmol/L MEHP处理组与0μmol/L组比较,继发坏死细胞比例明显增加(P<0.05)。结论一定浓度的DEHP及MEHP可诱导小鼠睾丸间质细胞早期凋亡。 Objective To explore the effects of DEHP and MEHP on apoptosis of leydig cells of mice. Methods Leydig cells were incubated with DEHP and MEHP at different dose groups (0, 1, 10 and 100 μmol/L)for 48 h respectively. Annexin V FITC--PI double staining with flow cytometry(FCM)was applied to detect leydig cells apoptosis in each group. Results The percentage of apoptosis cells increased significantly at 100 μmol/L DEHP group and 10 and 100 μmol/L MEHP groups in comparison with the control group(P〈0.05). The percentage of living cells decreased significantly(P〈0.05). The percentage of secondary necrotic cells increased distinctively in 10 and 100 μmol/L MEHP group compared with the control group(P〈0.05). Conclusions DEHP and MEHP in certain concentration might induce leydig cell apoptosis of mice.
