摘要
目的本文为找到镰孢菌(Fusarium concentricum)发酵产白僵菌素的适合发酵条件,在固液两种发酵方式下研究了影响白僵菌素合成的关键因素。方法对固态发酵培养基的含水量、液态发酵培养基组分以及培养条件进行了考察。结果发现固态发酵培养基含水量为1mL水/g玉米渣时白僵菌素产量最高。经初步优化后的液态发酵培养基为(g/L):淀粉40;酪蛋白胨10;NaNO31.5;MgSO4·7H2O 0.05;KH2PO44;KCl 1,摇床转速为200r/min。结论液态发酵条件下较高溶氧在一定程度上能促进白僵菌素的合成,但是剪切力对菌体生长及白僵菌素合成有不利影响。
Objective To identify the appropriate fermentation conditions for biosynthesis ofbeauvericin from Fusarium concentricum, the key factors affecting beauvericin production both in solid and liquid culture were studied. Method The water content of solid-state fermentation medium, the liquid fermentation medium components and culture conditions were investigated. Results Beauvericin reached the highest yield when the water content was lmL/g in solid medium. The optimal liquid culture media was composed of 40g/L starch, 10g/L casitone, 1.5g/L NaNO~, 4g/L KH2PO4, 0.05g/L MgSO4-7H20 and lg/L KCI, with the rotation speed at 200r/min. Conclusion In liquid medium,a relatively high concentration of dissolved oxygen can increase the production,while shear stress was disadvantageous to cell growth and synthesis ofbeauvericin.
