摘要
目的:采用RP-HPLC法测定宁夏固原地区4种地质条件下铁棒锤根、茎、叶及果实中乌头碱的含量并进行分析比较,为铁棒锤药材的标准化种植提供实验数据。方法:色谱柱ThermoODS-HYPER-SIL(4.6mm×250mm,5μm);DAD检测器;流动相0.1%乙二胺水溶液-甲醇-乙腈(37:39:24);流速1mL/min;柱温30℃。结果:测定了16批次样品,其中铁棒锤根部乌头碱的含量高,最高为固原地区黑城乡铁棒锤根中乌头碱的含量1.7494(mg/g);铁棒锤茎部中乌头碱的含量最低。结论:宁夏固原地区不同地质条件下的铁棒锤及其不同生长部位之间乌头碱的含量存在差异。所测定的乌头碱含量数据可作为宁夏铁棒锤药材标准化种植的实验依据。
Objective:To determine and compare the content of aconitine in 4 different parts of Aconitum Pendulum including roots, stems, leaves, fruits and from 4 regions of Ningxia by RP-HPLC which can be used to Aconitum flavum Hand. Mazz's standard planting. Methods: The chromatographic condition: Thermo ODS-2HYPERSIL(250mm × 4.6mm, 5um) column, DAD detection, the mobile phase consisted of 0.1 % ethanediamine water liquid, methanol and acetonitrile(37:39:24), with a flow rate of 1.0 mL · min -1 , at 30℃. Results : 16 samples were determined. The content of aconitine in roots is high. The highest content of aconitine in Aconitum Pendulum in Ningxia is in Heichen region. (1. 7494mg/g), the lowest is in stems. Conclusion:The content of aconitine in different part of Aconitum Pendulum from different region of NingXia is different. All the datas can be used to Aconitum flavum Hand. Mazz's standard planting.
