北柴胡镇痛有效物质组提取纯化工艺研究 被引量:6

摘要 目的:北柴胡为临床镇痛方剂的组成药味之一,经前期文献调研及药效学实验,确定北柴胡镇痛的有效物质组为多糖类成分,实验以多糖含量和出膏率为指标,探讨北柴胡多糖组分的提取纯化工艺条件。方法:采用L9(34)正交试验,以多糖含量和出膏率的综合指标为考察指标,对影响提取工艺的提取温度、溶剂量、提取次数、提取时间进行考察;以多糖纯度为指标,对影响纯化工艺的浓缩指数、醇浓度、静置时间进行考察。结果:确定柴胡多糖组分最佳提取条件为提取温度100℃,15倍量水,回流提取3次,每次1h;最佳纯化条件为提取液浓缩成1g/mL,80%的乙醇沉淀,静置6h。结论:确定北柴胡镇痛有效物质组的最佳提取纯化工艺条件,该优化工艺条件合理、可靠,在此工艺下得到的多糖组分纯度为30.69%。 Objective:Bupleurum chinense is one of the drugs in clinical analgesic prescriptions, according to early literature investi- gation and pharmacodynamics experiments, polysaccharides components were selected to be the effective substances. So we choose the polysaccharides contents and anointed rate as indexes to study the extraction and purification conditions for polysaccharides com- ponents from Bupleurum chinense. Methods:The L9(34) orthogonal design was used to investigate the influence of extracting tem- perature, solvent quantity, extracting times and extracting time on extraction process with the index of polysaccharides contents and anointed rate; then investigate the influence of concentration index, alcohol concentration and quiet place on purification process with the index of polysaccharide purity. Results : The optimal technological conditions were as follows., the temperature of extraction was 100℃, the ratio of water to sample was 15 : 1, and the time of extraction was 1 hour, extracting 1 time; and then the extract solution was condensed into 1 g/mL, precipitated with 80% alcohol, rest for 6 hours. Conclusion: The optimum conditions to extract and purify the polysaccharides components from Bupleurum chinense showed good stability and feasibility, and under this conditions, the purity of polysaccharides components can reach up to 30.69 %.
