NAC对大鼠I/R心肌细胞凋亡及bad、bak、bax的影响 被引量:2

摘要 采用末端脱氧核苷酸转移酶介导的带萤光的dUTP缺口末端标记 (TUNEL)法和免疫组化方法对N -乙酰基 -L -半胱氨酸 (NAC)对大鼠心肌缺血 4 5min再灌注 2 4h心肌细胞凋亡的影响及其对凋亡促进基因bad、bak和bax蛋白表达的调节进行研究。结果 :(1)NAC加IR组TUNEL法阳性心肌细胞核数量及阳性心肌细胞核占总心肌细胞核数的百分比均明显少于IR组(P <0 0 5) ;(2 )NAC加IR组表达bad、bak和bax蛋白阳性的心肌细胞数及阳性心肌细胞占心肌细胞总数的百分比均明显低于IR组 (P <0 0 5)。提示 :(1)NAC能够减少大鼠I/R心肌细胞凋亡 ;(2 )NAC能够下调I/R心肌细胞凋亡的多个凋亡促进基因的蛋白表达 ,这可能是NAC抑制I/R心肌细胞凋亡的机制之一。 This study is, using TUNEL and immunohistochemichal methed, to explore the effect of NAC on cardiac myocyte apoptosis and the regulation of it on bad, bak and bax during myocardial ischemia/reprfusion course in rat. The results showed:(1) The number of positive cardiac myocyte nucleus and the percentage of positive cardiac myocyte nucleus in NAC+IR group decreased significantly (P<0 05) compared with that of IR group respectively;(2)The number of protein bad, bak and bax positive cardiomyocyte and the percentage of protein bad, bak and bax positive cardiomyocyte in NAC+IR group were low (P<0 05) compared with that of IR group respectively. The results suggested that:(1)NAC could decrease cardiomyocyte apoptosis significantly. (2)Down-regulating the protein expression of bad, bak and bax in cardiomyocytes during I/R course might be one of the mechanisms of NAC.
