摘要
目的 研究感觉神经和运动神经来源的雪旺细胞 (Sc)表达低亲和力受体 (p75 )的情况。方法 结合酶消化、阿糖胞苷处理和S 10 0免疫荧光鉴定 ,从 5d龄SD乳鼠背根神经和股神经得到感觉和运动神经来源的Sc。 2种细胞分别进行普通培养基和添加高浓度白细胞介素 (IL) 1培养基的培养 ,检测不同时间Sc表面神经生长因子 p75的表达情况。 结果 感觉和运动Sc普通培养基组 p75受体表达方式明显不同 ,前者在第 5d迅速达到峰值 (10 86 .39± 5 35 .16 ) ,而后者在同期呈低水平(410 .5 0± 32 6 .5 5 ,P <0 .0 5 ) ;而添加IL 1组 2种细胞的表达基本一致 ,均在第 5d达到峰值(891.5 0± 40 6 .10、92 2 .32± 5 2 0 .2 5 ,P >0 .0 5 )后均迅速下降。结论 2种神经来源的Sc表达 p75受体存在差异 ,提示Sc在再生过程中存在感觉性和运动性的差异。
Objective\ To study the p75 expression of Schwann cells (Sc) derived from sensory and motor nerves. Methods\ Sc derived from dorsal root nerve or femoral nerve were taken in 5 day old SD rats and cultured in DMEM/F12, 10% FCS media with or without high dosage Interteukin (IL) 1 added. The expression of p75 was detected by using immunofluorescent method at indicated time points. Results\ Without IL 1 in media, the expression patterns of two groups of Sc were different. Sensory Sc displayed a peak expression at 5th day, and at the same time, the expression of motor Sc was relatively lower ( P <0.05). In the presence of IL 1 in media, the curves of p75 expression tended to be alike, and it reached the peak at 5th day ( P >0.05) and went downwards. Conclusion\ There is a biological difference between sensory and motor Sc, indicating a difference in sensory and motor exists in specific regeneration of peripheral nerves.
