摘要
目的 探讨骨巨细胞瘤化疗的可行性及确定生物可降解材料壳聚糖作为其治疗的药物载体的可能性。方法 体外培养骨巨细胞瘤标本 ,测定药物及壳聚糖的敏感性 ;将壳聚糖与化疗药制成缓释药粒 ,进行体外及体内释放试验。结果 骨巨细胞瘤对阿霉素 (ADM)、顺氨氯铂 (CDDP)中度敏感。单纯壳聚糖对骨巨细胞瘤在体外无抑制作用。体外及体内试验第 1dADM释放量较大 ,其后在较低水平维持相对稳定的缓慢释放达 8周以上。结论 骨巨细胞瘤可以进行化疗 ,但只适合缓释药粒的局部治疗。壳聚糖与敏感化疗药物制成的缓释制剂的局部应用具有局部浓度高、缓释时间长的优点。
Objective\ To investigate whether giant cell tumor (GCT) can be treated by chemical drug and whether biodegradable chitosan can be used as a drug carrier in treatment of GCT. Methods\ The specimens of GCT proved by pathology were cultured, then the sensitivity of the chemical drug and chitosan of the cells were tested. Chitosan and sensitive drug were mixed and formed drug delivery system (DDS), then the release tests of DDS were measured in vitro and in vivo. Results\ GCT was moderate sensitive to common dose of adriamycin (ADM) and cisplachitin (CDDP). The single chitosan had no inhibitory effect on GCT. The releaase dose of ADM from DDS at 1st day was maximum in vitro and in vivo release test, then the DDS could maintain sustained and show release over 8 weeks. Conclusion\ GCT could be treated by chemical drug, but it is only suitable for local treatment by DDS.
