肝硬化门静脉高压症患者肝组织α_1肾上腺素受体亚型的mRNA表达

Expression of α_1 adrenoceptor subtypes mRNA in liver tissues of cirrhotic patients with portal hypertension

目的 探讨肝硬化门静脉高压症患者肝组织α1肾上腺素受体 (α1AR)亚型mRNA的表达情况。方法 应用半定量逆转录聚合酶链反应法检测 12例乙型肝炎后肝硬化肝组织和 15例正常肝脏组织中α1AR亚型mRNA的表达。同时逆转录扩增α1AR亚型特异性片段和内参片段 ,以二者的积分吸光度比值作为该受体亚型的mRNA相对表达量。结果 肝脏组织中α1a、α1bAR亚型的相对表达量对照组 (0 .86± 0 .38、0 .2 3± 0 .10 )均显著高于肝硬化组 (0 .2 6± 0 .12、0 .0 3± 0 .0 1,P <0 .0 1)。对照组及肝硬化组肝组织中α1a亚型的表达量均显著高于α1b亚型 (P <0 .0 1)。两组肝组织中均无α1d亚型的表达。结论 肝硬化肝组织中α1a和α1bAR亚型基因表达减少是造成肝硬化时肝脏α1AR蛋白减少的重要原因 ,对门静脉高压症的形成和维持可能起重要作用。

Objective\ To investigate the expression of mRNA of α 1 adrenoceptor (α 1AR) in liver tissues of cirrhotic patients with portal hypertension. Methods\ Semi quantitative reverse transcription and polymerase chain reaction (sqRT PCR) was used to detect the expression of mRNA of α 1 AR subtypes in liver tissues in 12 hepatitis B virus related cirrhotic patients and 15 controls. In these patients routine liver biopsies were performed. Special α 1 AR subtype product and internal standard GAPDH product were both amplified by RT PCR. The ratio of their integral optical density (IA) was calculated as the relative expression of the α 1 AR subtypes.Results\ The relative mRNA expression of α 1a and α 1b AR subtype in liver tissues of cirrhotic patients (0.26±0.12,0.03±0.01) was significantly lower than that of controls (0.86±0.38, 0.23±0.10, P <0.01). Neither cirrhotic patients nor controls showed α 1d AR subtype in liver tissues. The relative mRNA expression of α 1a AR subtype was significantly higher than that of α 1b AR subtype in liver tissues of both cirrhotic patients and controls ( P <0.01). Conclusion\ The decreased expression of mRNA of α 1a and α 1b AR may underlie the decrease of α 1 adrenoceptor proteins often seen in the liver tissues of cirrhotic patients, and may play an important role in the pathogenesis and maintenance of portal hypertension.
