摘要
为研究艾叶多糖对小鼠腹腔巨噬细胞吞噬功能及对巨噬细胞生成NO的影响,制备艾叶多糖,灌洗法提取小鼠腹腔巨噬细胞,纯化后DMEM培养液培养,用不同浓度的艾叶多糖与巨噬细胞共孵育,进行墨汁吞噬实验。显微镜观察巨噬细胞吞噬墨汁的情况,酶标仪定量检测巨噬细胞内墨汁的吸光度值,并计算吞噬率;用标准曲线法测定孵育96 h后的巨噬细胞的NO生成量。结果显示:艾叶多糖能增强巨噬细胞吞噬墨汁以及生成NO的能力,在实验剂量范围内均有剂量-效应关系。研究表明艾叶多糖对体外培养巨噬细胞具有免疫调节功能。
Objective : to investigate the effect of artemisia argyi polysaccharidc on the phagocytosis and NO gen-eration of abdominal macrophage in mice. Methods: artemisia argyi polysaccharide was prepared, and ab-dominal macrophages in mice were isolated, purified and cultured in DMEM medium. After incubation of macrophages with different concentration of artemisia argyi polysaccharide, ink phagocytosis test was carried out to qualitatively observe and quantitatively measure macrophages^ability to phagocytose ink. The standard curve was c^ated with sodium nitrite to indirectly measure the NO generation of macrophages after incubation for 96h. Results : wittfin a series of exposure dosages, there existed relationship of dose - effect of artemisia argyi polysaccharide to macrophages's ability to phagocytose ink and to secrete NO. Conclusion: artemisia arg-yi polysaccharide has immunomodulating activity to the macrophages in vitro.
