摘要
目的研究血吸虫重组BCG-Sj26GST疫苗对小鼠T淋巴细胞亚群的影响。方法实验一采用106和108CFU疫苗皮下接种BALB/c小鼠,接种后8WK用日本血吸虫尾蚴进行攻击感染,感染6wk剖杀小鼠,取脾制备脾细胞,同时设有PBS对照组。实验二分别用106CFU疫苗皮下和静脉注射免疫小鼠,分别于免疫后0、4、8、10、14和16wk各剖杀4只,分离脾脏,用FACsort流式细胞仪分析T细胞CD4+和CD8+亚群百分比。结果实验一疫苗免疫络小鼠经尾蚴攻击后脾T细胞CD4+亚群明显升高,CD8+亚群缓慢增加。实验二动态观察发现在皮下和静脉注射组,CD4+亚群分别于免疫后10和18WK显著增加;皮下注射组CD8+亚群在整个观察期间均无明显变化,静脉注射组则于免疫后4~16wk有轻度增加。结论T细胞CD4+亚群可能在疫苗保护性免疫中起重要作用。
Aim To study the effects on T lymphocyte subsets in mice of mice immunized with recombinant BCGSj26GST.Methods In the first experiment, BALB/c mice were immunized subcutaneously by 106 and 108 CFU BCG-Sj26GST respectively.ALL the mice were artificially challenged with cercariae of Schistosoma japonicum on 8weeks after immunization,six after challenge, the mice were killed, and the spleens were removed, cells were prepared seperately and were analysed by immunofluorescence with conjugated monoclonal antibodies in a FACsort cytofluorimeter at 523 nm,PBS treated mice were served as control.In the second experiment,after immunized subcutanceously or intravenlusly by 106 CFU vaccine,4 mice were ranndomly killed to separate spleens on 10 wk, 14wh and 16wk after immunization, the percentage of CD1+ and CD8+ subsets was analysed as above.Results In the first experiment, CD4+ suhas incresaed remarkably, but CD8+ be did slightly by immunization with the vaccine against challenge with S.j.Cercariae, in the second experiment, the dynamic observation showed that in the subcutaneous and intravenous roup, CD4+ subsets increased obviously since 10wk and 8wk respectively,CD8+ subsets had no obvious change over 16 weeks of observation,in the subcutaneous group but the CD8+ subsets rose lightly on 4-16wk in the intravenous group.Conclusion On the basis of this study it's suspected that CD4+ subsets might play an important role in the protective immunity induced by the vaccine
