抑癌基因PTEN与凋亡抑制因子Livin在乳腺癌中的表达及相关性 被引量:2

摘要 目的探讨抑癌基因PTEN与凋亡抑制因子Livin在乳腺癌中的表达及两者之间的关系,为乳腺癌的诊断及临床治疗提供依据。方法采用免疫组织化学法分别检测50例乳腺癌、20例癌旁乳腺组织及10例正常乳腺组织中PTEN及Livin的表达。结果 PTEN蛋白在正常乳腺组织、癌旁乳腺组织及乳腺癌组织中的高表达率分别为100.0%、90.0%及40.0%,前两者与后者比较,差异有统计学意义(P<0.05)。乳腺癌组织中PTEN的表达与组织学分级、临床分期及腋下淋巴结转移均呈显著相关(P<0.05),PTEN的表达与年龄、肿瘤大小及远处转移无显著相关(P>0.05)。Livin蛋白在正常乳腺组织、癌旁乳腺组织及乳腺癌组织中的高表达率分别为40.0%及58.0%,前两者与后者比较,差异有统计学意义(P<0.05)。乳腺癌组织中Livin的表达与组织学分级、临床分期及腋下淋巴结转移均显著相关(P<0.05),Livin的表达与年龄、肿瘤大小及远处转移无显著相关(P>0.05)。乳腺癌组织中Livin表现高表达,而PTEN则表现低表达,两者之间呈负相关(r=-0.298,P<0.05)。结论 Livin的异常高表达和PTEN的低表达可能与乳腺癌的发生发展密切相关,Livin和PTEN在乳腺癌组织中的表达呈负相关性,提示Livin和PTEN的表达可能成为判断乳腺癌侵袭能力的重要基因指标。 Objective To investigate the expressions of tumor suppressor-PTEN and apoptosis suppressor-Livin in breast cancer and the correlation between them,and to provide theoretical bases for the diagnosis and treatment of breast cancer.Methods The expressions of PTEN and Livin were detected by immunohistochemistry in 50 cases of breast cancer tissues,20 cases of adjacent-carcinoma tissues and 10 cases of normal breast tissues.Results The expression levels of PTEN in normal breast tissues,corresponding adjacent-carcinoma tissues and breast cancer tissues were 100%,90%,40%,respectively,and there were significant differences in the expression of PTEN between the breast cancer tissues and the other two kinds of tissues (P〈0.05).The expression of Livin in breast cancer was significantly correlated with the histological grade,clinical staging and axillary lymph node metastases (P〈0.05),however,which was not related with patients’ age,tumor’s size and distant metastasis (P〉0.05).The Livin was highly expressed in breast cancer tissues,however,PTEN was lowly expressed,and the expression of Livin was negatively correlated with that of PTEN (r=-0.298,P〈0.05).Conclusion The high expression of Livin and the low expression of PTEN may play an important role in the pathogenesis and progression of breast cancer.The expressions of Livin are negatively correlated with those of PTEN,which suggests that the expression levels of Livin and PTEN can be used as the important indexes for evaluating the invasive ability of breast cancer.
