摘要
目的检测TNF-α对人肝癌细胞系中CD15、CD15s和ST3GaL糖基转移酶含量及细胞侵袭能力的影响。方法应用Western blot方法检测TNF-α作用前后人原发性肝癌细胞系HepG-2和人高转移肝癌细胞系SMMC-7721中CD15,CD15s和ST3GaL糖基转移酶含量的变化,Transwell检测TNF-α作用前后HepG-2、SMMC-7721细胞侵袭力的变化情况。结果 TNF-α作用后,HepG-2细胞系CD15含量较对照组下降(P<0.05),CD15s含量较对照组明显上调(P<0.05),ST3GaL糖基转移酶家族蛋白(6种)中ST3GaL-Ⅱ和ST3GaL-Ⅳ表达较对照组明显上调(P<0.05),其他4种ST3Gal-Ⅰ、ST3Gal-Ⅲ、ST3Gal-Ⅴ和ST3Gal-Ⅵ的表达无明显变化(P>0.05),细胞侵袭力提高(P<0.05)。SMMC-7721细胞系CD15、CD15s含量较对照组无明显改变(P>0.05),细胞侵袭力无显著变化(P>0.05)。结论在人原发性肝癌细胞系HepG-2中,TNF-α可能通过影响CD15、CD15s、糖基转移酶ST3GaL-Ⅱ、ST3GaL-Ⅳ的含量促进细胞侵袭能力。
Objective To detect the expressions of CD15,CD15s and ST3GaL in human primary liver cancer cell lines HepG-2 and human high metastatic liver cancer cell line SMMC-7721 treated with TNF-α,analyze the relationship among CD15,CD15s and ST3GaL contents,and study the effect of TNF-α on the invasion of HepG-2 and SMMC-7721 cells.Methods HepG-2 and SMMC-7721 cells were treated with TNF-α for 24 h and 48 h,respectively,and the expression contents of CD15,CD15s and ST3GaL were detected by Western blot.The effect of TNF-α on the invasion of liver cancer cell lines HepG-2 and SMMC-7721 was evaluated by Transwell assay.Results Western blot showed that CD15 was down-regulated and CD15s was up-regulated in primary liver cancer HepG-2 cells with TNF-α treatment(P 0.05).Moreover,ST3GaL-Ⅱ and ST3GaL-Ⅳ were up-regulated simultaneously in primary liver cancer HepG-2 cells treated with TNF-α(P 0.05),but other ST3GaL glycosyltransferase family members:ST3Gal-Ⅰ,ST3Gal-Ⅲ,ST3Gal-Ⅴand ST3Gal-Ⅵ had no obvious change(P 0.05).After treatment of TNF-α,the HepG-2 cell invasion capability was enhanced.However,there was no significant change of CD15 and CD15s contents in SMMC-7721 under TNF-α treatment(P 0.05).The SMMC-7721 cell invasion capability had no significant change.Conclusion TNF-α modulates the content of CD15,CD15s and the expressions of ST3GaL-Ⅱ and ST3GaL-Ⅳ in human primary liver cancer cell line HepG-2,and affects their invasion in vitro.
