摘要
目的:研究药用真菌杨树菇活性组分对荷瘤小鼠的生殖毒性影响。方法:使用HP-20大孔吸附柱分离杨树菇抗肿瘤活性成分Yt成Yp、Ys,构建荷H22小鼠模型,瘤内注射活性组分(2.5mg/kgYp、2.5mg/kgYs、5mg/kgYt),通过小鼠体重变化、雄鼠睾丸组织切片、精子活力及活率来研究活性组分的生殖毒性作用。结果:与对照组相比,瘤内注射Yp、Ys、Yt对荷瘤小鼠的体重及精子活力无显著性影响,睾丸组织也无明显病理变化,但显著地抑制精子活率。结论:杨树菇抗肿瘤活性成分可能会有雄性生殖毒性,需要进一步的分离纯化促进其合理利用。
Objective The aim of this study was to investigate the reproductive toxicity of bioactive components from medicinal fungus Agrocybe aegerita. Methods The antitumor components Yt from Agrocybe aegerita was seperated to Yp and Ys using HP-20. Yp (2.5 mg/kg), Ys (2.5 mg/kg), and Yt (5 mg/kg) were injected intratumorally on H22-bearing mouse models, and the reproductive toxicity of the components was measured by testing animal weight, the histological analysis of testis, the morality and living rate of the sperm. Results Yp, Ys and Yt did not induce significant changes in mouse weight, sperm motality and histological result of testis compared to the control group, but could inhibit the sperm living rate significantly. Conclusion The antitumor components from Agrocybe aegerita might have reproductive toxicity, and need to be purified for the further application.
