Effects of isoflurane on cell cycle and apoptosis in PC12 cells

摘要 目的探讨异氟醚对PCI2细胞株细胞周期与凋亡的影响。方法经神经生长因子孵育7d的神经元样PCI2细胞,培养于25crn2培养瓶,采用随机数字表法,将其随机分为2组(n=6):对照组(c组)和异氟醚组(I组)。C组正常培养,I组用1.2%异氟醚处理12h。采用倒置相差光学显微镜观察细胞形态;收集细胞,采用流式细胞术测定细胞凋亡率、细胞周期、线粒体膜电位(MMP)及胞浆钙离子浓度([Ca2+].)。结果与C组比较,I组G0/G1期细胞比例减少,S期和G2/M期细胞比例增加,细胞凋亡率增加,MMP降低,[Ca2+].增加(P〈0.05)。l组细胞形态发生损伤变化。结论1.2%异氟醚处理神经元样PCI2细胞12h,可异常启动细胞周期,致细胞发生凋亡。 Objective To investigate the effects of isoflurane on the cell cycle and apoptosis in PCI2 cells. Methods The neuronal PC12 cells were cultured for 7 d with nerve growth factor in vitro. The cells were cultured in 25 cm2 culture flask and randomly divided into 2 groups (n = 6 each) : control group (group C) and isoflurane group (group I). The neuronal PC12 cells were exposed to 1.2% isoflurane for 12 h in group I. The cell morphology was examined by light microscopy. The apoptotic rate, cell cycle progression, mitochondrial mem- brane potential (MMP) and intracellular Ca2. concentrations ( [ Ca2 + ] i ) were assessed by flow eytometry. Results Compared with group C, the cell morphology was changed, the proportion of the cells in G0/G1 phase and MMP were signiilcantly decreased, and the proportion of the cells in G2/M phase, apoptotic rate and [ Ca2+ ]i were sig- nificantly increased in group I ( P 〈 0.05) . Conclusion Exposure of PC12 cells to 1.2% isoflurane for 12 h can activate the cell cycle abnormally and result in cell apoptosis.
