摘要
目的:建立齿瓣石斛HPLC特征图谱。方法:采用Zorbax SB-Aq色谱柱,流动相为乙腈-0.2%甲酸水溶液(梯度洗脱),流速为1.0 mL.min-1,检测波长分别为270 nm及340 nm,柱温为35℃。结果:在270 nm检测波长条件下,齿瓣石斛标示出34个特征共有峰,10批样品的相似度为0.919~0.968;在340 nm检测波长条件下,专属性地检出10个黄酮类特征共有峰,10批样品的相似度为0.882~0.981。齿瓣石斛鲜品检出与齿瓣石斛基本一致的特征共有峰,相似度大于0.9。部分市售齿瓣石斛不同程度掺假兜唇石斛。结论:方法准确可靠,重复性好,为齿瓣石斛的质量控制提供了方法依据。
Objective:To determine the spectrum characteristics of Dendrobium devonianum by HPLC. Method: HPLC was used with the Zorbax SB -Aq column, the acetonitrile -0. 2% methanoic acid solution (gradient elution) was employed as a mobile phase, flow rate was 1.0 mL · min- 1, detection wavelength was 270 nm and 340 nm, and column temperature was 35 ℃. Results: A total of 34 common characteristic peaks in 270 nm were determined for Dendrobium devonianum, and the similarities among 10 batches were 0. 919 -0. 968. 10 common characteristic peaks(flavonoids) in 340 nm were determined and the similarities of 10 batches were 0. 882 -0. 981. Fresh Den- drobium devonianum had the same characteristic spectrum as Dendrobium devonianum, the similarities between the fresh sample and the species were higher than 0.9. Dendrobium devonianum from the market often falsified with dif- ferent degree of adulterated Dendrobium aphyllum. Conclusion:The method is accurate, reliable, and with good re- producibility, which provides a reference for the quality control of Dendrobium devonianum.
